槲皮素对仔猪生长性能、肠道抗氧化及免疫相关基因表达的影响Effects of quercetin on growth performance and intestinal antioxidant capacity and immune related gene expression in piglets

摘要(Abstract):
【目的】通过分析不同含量槲皮素对仔猪生长性能、肠道抗氧化指标、炎症细胞因子分泌及紧密连接蛋白基因表达的影响,探究槲皮素在改善仔猪应激综合征的作用。【方法】选取216头健康、初始体质量为(9.5±0.5)kg(杜×长×大)仔猪,随机分为3组(对照组、试验1组和试验2组),分别饲喂基础日粮、基础日粮+200 mg·kg~(-1)槲皮素日粮和基础日粮+400 mg·kg~(-1)槲皮素日粮,每组6个重复,每个重复12头仔猪(公、母各6头),试验期为15 d。【结果】1)与对照组相比,试验1组和试验2组均提高了仔猪平均日增重,显著降低了腹泻率。2)与对照组相比,试验1组和试验2组显著提高了仔猪空肠黏膜谷胱甘肽过氧化物酶(glutathione peroxidase, GPx)、超氧化物歧化酶(superoxide dismutase, SOD)和过氧化氢酶(catalase, CAT)活性,显著降低脂质过氧化物丙二醛(malondialdehyde, MDA)含量。其中,GPx、SOD和CAT活性随着槲皮素添加含量提高而显著增加。3)与对照组相比,试验1组和试验2组显著降低了空肠黏膜中白细胞介素2(interleukin-2,IL-2)、白细胞介素1β(interleukin-1β,IL-1β)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)水平,显著提高了白细胞介素10(interleukin-10,IL-10)水平;与其他组相比,试验2组分泌型免疫球蛋白A(secretory immunoglobulin A,sIgA)水平显著增加。4)与对照组相比,试验1组和试验2组显著提高了仔猪空肠黏膜中紧密连接蛋白基因Claudin-1、Occludin和ZO-2mRNA相对表达量,并且试验2组显著提高了ZO-1 mRNA相对表达量;与试验1组相比,试验2组显著提高仔猪空肠黏膜中紧密连接蛋白基因Claudin-1和Occludin mRNA相对表达量。【结论】日粮添加槲皮素可以降低断奶后仔猪腹泻率,改善生长性能,增强肠道抗氧化功能,提高抗炎因子分泌,抑制促炎因子分泌,促进肠道紧密连接蛋白相关基因表达。
