血清单稀释 ELISA 检测鸡 ILT 抗体的初步研究An ELISA for Detecting Antibody in Chicken Serum to ILTV in a Single Serum Dilution

摘要(Abstract)：
应用鸡胚皮肤细胞培养传染性喉气管炎(ILT)病毒,经超速离心浓缩后制成抗原,兔抗鸡IgG 经辣根过氧化物酶标记,制成酶标抗体,进行酶联免疫吸附试验(ELISA),确定了该实验的最适条件。血清单稀释度(1:80)的光密度值(OD 值)与终点法所测定的ELISA 滴度(ET)的自然对数间有良好的线性关系:Ln(ET)=5.80(OD)+3.76(r=0.94>r0.01),说明可利用血清的单稀释度测定鸡血清中ILT 抗体,同终点法相比,单稀释ELISA 可将检查效率提高8～10倍,是一种快速、敏感的实用方法。本实验还研究了人工感染、疫苗免疫后鸡血清中的抗体消长情况,比较了单稀释ELISA、血清中和实验(SN)、琼脂扩散沉淀试验(AGP)的特点。
