基于多序列比对的甜瓜SSR标记开发及应用Development of SSR markers based on multiple sequence alignment and their applications in melon

摘要(Abstract):
采用多序列比对的方法,从甜瓜EST序列中发现约1 100个SSR差异位点,并随机挑选50个位点设计成SSR引物,对47份遗传背景不同的甜瓜种质进行PCR扩增。结果表明,41对引物可产生清晰的带谱,有效扩增率达82.00%,且均具有多态性。共检测到130个等位基因,平均3.17个。Na、Ne、Ho、He、PIC等参数表明,这些标记具有一定程度的多态性。本研究开发的SSR标记可用于甜瓜的遗传多样性和群体结构分析,基于多序列比对的标记开发方法亦可在基因组信息丰富的物种中应用。
